1447 Anti-senescence effect of extracellular vesicle from hyaluronic acid primed induced mesenchymal stem cells on aged human dermal fibroblast

Skin aging, represented by wrinkles, is caused a decrease in the dermal components such as collagen, elastin, and fibroblast. Extracellular vesicles (EVs) mediate cell-to-cell communication via cargo biomolecules cell-free manner. In previous studies, we produced mesenchymal stem cell (MSC)-derived cargo-regulated EVs through various priming methods reported that these affect progression of diseases. Here report effect derived from HA-primed iMSCs (HA-iMSC-EVs) on aged human fibroblast (HDF). HA-iMSC-EVs increased proliferation expression HA receptor, CD44, insulin-like growth factor 1 receptor (IGF1R) HDFs cultured for prolonged period. addition, enhanced accumulation extracellular elastin fibers HDFs. Also, decreased senescence associated-beta-galactosidase (SA-β-gal) activity, while itself did not reduce SA-β-gal activity. shift toward S+G2/M phase cycle was observed HA-iMSC-EVs-treated Consistently, similar outcome HDF’s induced glucocorticoid steroids cultivation. detail, dexamethasone reduced type collagen (COL1A1) ELN concentration-dependent manner HDF without affecting proliferation. However, restored COL1A1, IGF1R Cyclin D increased, suggesting also promotes HDF. Conclusively, our results suggest have potential to improve skin aging stimulating restoring production tissue.
